The main route into Plymouth is the A38 Devon Express way which is a continuation of the M5. From the North West use M6 southbound then M5 for the South West
From London, Thames Valley and Swindon use the M4 which joins the M5 at Bristol.
Alternatively the A303 (via M3) which cuts through Salisbury Plain passing Stone Henge is the more scenic route which joins the M5 at Honiton nr Exeter.
By Train:
